So she has been popping, idling rough and mild backfires.   Vacuum lines are near new, tightened all the 18 clamps, have new intake orings.  So  I decided to take apart and clean the carbs.  Great You tube video tells you most of how to do it.
One of the interesting things it says to do is count while closing the needle valve to record number of turns until it seats. Use that adjustment for reassembly of pilot screws.
So 5 out of 6 of them are sitting at 1.75 to 2.1/2,2 turns in till it seats.  However I find one hits the seat at 1/4 turn! Crazy.   So the question is, should I reassemble like the video says taking the one guy  down to 1/4 turn out, or go with gut feel and bring it out to 1 3/4 turn to match the other 5?

Thanks. Its been an interesting project while I am at home with a recovering - from - surgery wife...Thanks

Was it the front left cylinder?
If so I have seen that before on a mal adjusted high altitude bike. (That ran bad)

I reset it to standard, in my case 2.25 turns, everything was fine.

Usually the factory setting is 1.75. The one at .25 could have been done by someone trying to set them with a tach.
If the low jets are 35s then open them to 2.25, all of them.

Thanks all.  The jets are the 35's. It was #4 carb as the culprit.

Set all of the mixture screw at 2 1/4 turns out.
