I have an interested tail/brake/turn signal on my fat lady the tail light works,  the turn signals work but when I apply the brakes nothing lights up.  What should I check and in what order?

Are you using the rear lever, front brake lever or both?

If it is the rear brakes I'd check to make sure the switch is adjusted correclty at the brake lever.   If you look closely, you will see the little switch.

Very easy task.   If the front is all you are using, I'd look for a blown fuse.

Used both the front brake lever and the rear brake foot pedal and the brake is not lighting up.

You need to check for pwr. at the brk. input to your rbt module, most modules pwr. up from the tail lamp circ.
If you have a schematic for the module it would make this easier.

Pull the bulb out and eye ball it to the sun to see if one of the little wire strands is burned out.

I'll assume that any new fixture would be LED? Is this an additional light fixture? If so, does the original brake light illuminate? If so you have the brake wire spliced into the wrong wire. Wrong wire would be the culprit even if this is a stand alone fixture.

doniholcomb,

A little more information would be really helpful.  This is presumably an aftermarket set up.

Did it ever work properly?

If it is a new install (or even an old installation), who is the manufacturer?

If you wired it yourself, can you provide details (or a diagram) of what you did?

It is really hard to diagnose a lighting system with no idea of how it is wired.

As an aside, here is the ugly 4 relay, 2 diode system I made for my rear lights:
